Roche, Nurix Partner on BTK Degrader Bexobrutideg

Roche BTK degrader

By VETTAPHARMA reporter – Derek Roche: Roche has entered an exclusive licensing and collaboration agreement with Nurix Therapeutics to co-develop and co-commercialize bexobrutideg (NX-5948), an investigational Bruton’s Tyrosine Kinase (BTK) degrader being advanced across malignant hematology, immunology, and neurology indications.

The collaboration strengthens Roche’s hematology portfolio while expanding its pipeline into chronic spontaneous urticaria (CSU) and multiple sclerosis (MS).

Bexobrutideg is an oral, brain-penetrant targeted protein degrader designed to eliminate BTK protein rather than simply inhibit its activity, potentially overcoming resistance mechanisms associated with current BTK inhibitors.

The companies plan to advance bexobrutideg through a broad clinical development program spanning B-cell malignancies, immunology, and neurology. Roche noted that a Phase 3 trial is expected to begin in summer 2026 in second-line chronic lymphocytic leukemia (CLL), where patients frequently experience disease progression due to acquired resistance mutations, incomplete pathway suppression, or treatment intolerance.

Available clinical data suggest bexobrutideg may offer higher efficacy and improved tolerability compared with established BTK-targeted therapies. By removing both the kinase activity and scaffolding function of BTK, the investigational therapy may address treatment-emergent resistance mechanisms while potentially extending BTK targeting into immune-mediated and neurological diseases.

The agreement includes an upfront payment of $700 million to Nurix, with the biotechnology company eligible for development, regulatory, and commercial milestone payments that could bring the total transaction value to up to $2.3 billion. Development costs will be shared, with Roche funding 60% and Nurix 40%.

The companies will equally share profits and losses from U.S. commercialization and will co-commercialize bexobrutideg across all approved indications in the United States. Outside the U.S., Roche will lead commercialization efforts while Nurix will receive tiered royalties ranging from the low- to high-teens.

Roche highlighted the commercial significance of BTK-targeted therapies, citing projections that the combined non-Hodgkin lymphoma (NHL) and CLL markets could reach $41 billion by 2031, with BTK inhibitors expected to account for approximately $19 billion in sales. The CLL market alone is projected to grow from $12 billion in 2024 to $16 billion by 2035.

The transaction remains subject to customary closing conditions, including antitrust review, and is expected to close in the third quarter of 2026.

Quick FAQs

1. What Is Bexobrutideg?

Bexobrutideg (NX-5948) is an investigational oral, brain-penetrant BTK degrader designed to selectively eliminate BTK protein from cells and potentially overcome resistance associated with conventional BTK inhibitors.

2. What Happened?

Roche and Nurix Therapeutics announced a global licensing and collaboration agreement to co-develop and co-commercialize bexobrutideg across hematology, immunology, and neurology indications.

3. What Is The Clinical Development Program?

The collaboration includes development across B-cell malignancies, chronic spontaneous urticaria, and multiple sclerosis. A Phase 3 trial in second-line CLL is planned to begin in summer 2026.

4. What Is The Disease Condition?

The lead indication is chronic lymphocytic leukemia, a common adult blood cancer in which patients frequently require additional treatment following disease recurrence. The program also targets CSU and MS.

5. What Were The Key Results?

Clinical data generated to date suggest bexobrutideg has the potential to deliver higher efficacy, improved tolerability, and activity against resistance mechanisms observed with existing BTK inhibitors. A Phase 3 study is planned for second-line CLL.

6. Why Is This Important?

The collaboration gives Roche access to a potential best-in-class BTK degrader while expanding development opportunities beyond oncology into immunology and neurology. The deal also supports advancement of a program targeting large and growing NHL and CLL markets.
