Sanofi’s Cenrifki (tolebrutinib) has been recommended for approval by the Committee for Medicinal Products for Human Use (CHMP) in the European Union for the treatment of secondary progressive multiple sclerosis (SPMS) without relapses.
Tolebrutinib is an oral, brain-penetrant Bruton’s tyrosine kinase (BTK) inhibitor, designed to target disease-driving mechanisms within the central nervous system.
The CHMP recommendation is based on clinical data demonstrating that tolebrutinib can impact disability progression in patients with non-relapsing SPMS, a population with limited treatment options and high unmet medical need.
Secondary progressive multiple sclerosis represents a later stage of the disease characterized by gradual worsening of disability independent of relapses, making effective treatment particularly challenging.
If approved by the European Commission, tolebrutinib could become one of the first therapies specifically addressing progression independent of relapse activity, offering a new therapeutic approach for patients with this form of multiple sclerosis.
From a commercial perspective, the potential approval strengthens Sanofi’s neurology pipeline and positions tolebrutinib within a high-value multiple sclerosis market. Expanding into progressive forms of the disease could provide meaningful revenue opportunities, given the limited competition and significant unmet need in this segment.
